Replicating Heart Stem Cells

57 year old Ted Barnes knew that his body was sounding an alarm, and guessed he was experiencing the symptoms of a heart attack. "I was feeling a tingling in my fingertips and it was kind of an unusual feeling. Then I had this attack of sweating that I never had before, which left me feeling nauseous."


Ted Barnes drove himself straight to the emergency room where doctors determined he had had a heart attack. After closer examination, Ted was diagnosed with cardiomyopathy. It's when the heart muscle becomes inflamed and doesn't work as well as it should. Unfortunately, Ted's condition has worsened. "I'm on the heart transplant list, and that's a scary proposition." says Ted.


Researchers at Johns Hopkins University are now working towards perfecting stem cell biology of the heart, so that one day, patients like Ted can actually repair their own heart when it gets damaged from a heart attack or as a result of heart failure, using their own tissue. "The beautiful thing about these cells is that they are actually specialized to become heart muscle. So our hope is that by growing these in a dish and then putting them back in the same patients, they not only grow healthy heart muscle but their own heart muscle, completely identical to the host that it came from." explains Dr. Eduardo Marban, Johns Hopkins Heart Institute.


By using a person's own adult stem cells instead of those from another donor, there would be no risk of triggering an immune response that could cause rejection of the heart stem cells. Johns Hopkins scientists have developed a method for collecting heart stem cells from small amounts of biopsied heart tissue and growing the cells in the lab to get more. "We would take those biopsies, grow them in a dish and bring the patient back some weeks later when we have enough cells." explains Dr. Marban. The resulting clusters, called cardiospheres contain cells that retain the ability to regenerate themselves and to develop into more specialized heart cells that can conduct electrical currents and contract like heart muscle should. "It works so dramatically in animal models that we have reason to hope that it will be similarly useful in humans." says Dr. Marban. Ted Barnes would love to be the first success story. "I'm only hoping that the research would occur in my lifetime." says Ted.


According to Dr. Marban, his hope is that human trials on heart stem cells will begin within the next 2 years.
